1、本课题对影响阿司匹林微囊缓释性的影响因素进行研究,选取阿司匹林作为芯材,明胶与海藻酸钠为壁材,采用复凝聚法制备阿司匹林-明胶-海藻酸钠微囊。在体外释放实验下,测定累积释放百分比(Q%)并且作为微囊缓释性能评价指标。在单因素试验下考察不同的固化温度、海藻酸钠PH值、明胶浓度、交联剂用量等因素对微囊缓释性的影响;再通过Box-Behnken响应面法对上述因素进行分析。实验表明:影响载药微囊缓释性大小因素为:海藻酸钠PH值明胶浓度固化温度交联剂用量;得到最佳制备工艺条件:固化温度为40. 15,海藻酸钠PH值为6. 50,明胶浓度为2. 04%,交联剂用量为23. 72mL,累积释放百分比(Q%)为
2、69. 95%,该条件下制备的微囊具有良好的缓释性能。关键词:明胶;海藻酸钠;阿司匹林;微囊AbstractAbstractAbstractIn this paper, the factors affecting the sustained release of aspirin microcapsuleswere studied. Aspirin was selected as core material, gelatin and sodium alginate wereused as wall materials, and aspirin-gelatin-sodium alginate mi

In vitro release test, the cumulative releasepercentage (Q%) was determined and used as an index to evaluate the sustained releaseperformance of microcapsules. The effects of curing temperature, pH value of sodiumalginate, gelatin concentratio
4、n and dosage of cross-linking agent on the sustainedrelease of microcapsules were investigated by single factor test, and the above factorswere analyzed by Box-Behnken response surface method. 7、示为阿司匹林化学结构示意图,由上图分析可知:本品粉末为白色结晶性,带有部分醋酸气味田。该结构式中具有酯键,遇湿润气易缓慢水杨酸和乙酸,应当干燥条件下密封存放。木品溶于水(其水溶液显酸性,PKa=3.5),易溶于乙醇、乙醛等。本品在沸水中易水解,试验时应当控制温度。因Hoffman通过乙酰化反应,把水杨酸制备成阿司匹林。距今以来,广泛应用到临床领域(治疗感冒、伤风、解热、镇痛、抗炎等)已有百年历史,一直被人们称之为“神药”。1.2 阿司匹林的药理作用解热作用:与下丘脑的体温调节不平衡有关。由于下丘脑是维持正常体温调节中枢。在人体正常体温条件下,下丘脑的体温调节中枢产热和散热之间维持着动态平衡状态
